You need to open the files as root. The best way would be to:

su, <password>, then start an editor such as kate, gedit or openoffice, at the command line. Then you can simply use the editor to open the files, modify them, and then save.

The files are self documented, so it should be easy. To update clamav, you need root acccess as you are updating a system file. To do this:

su
<password>
freshclam

The most important change is the source for updates, for example:

# Uncomment the following line and replace XY with your country
# code. See http://www.iana.org/cctld/cctld-whois.htm for the full list.
# Default: There is no default, which results in an error when running freshclam
DatabaseMirror db.au.clamav.net

I'd consider installing klamav. This uses the clamav packages, but uses a graphical interface for all tasks. Also, since it stores virus definitions in /home/xxxx, you do not need to use root access for updates.
