Does the grub that comes with the latest alpha support booting from an ext4 root partition? That is, do I still need to create a seperate /boot?

Yes, it does support ext4 on /boot.

I've installed the alpha without /boot partition and / and /home where both ext4. So, F12 supports booting from ext4.
